LY-411575 is a potent γ-secretase inhibitor as assessed by inhibition of Aβ production (IC50, 0.078 and 0.082 nM in membrane- and cell-based γ-secretase assays). LY411575 is a selective, cell permeable, small molecule gamma secretase inhibitor which blocks Notch activation. Notch signaling is a pathway known to regulate both differentiation and proliferation of cells in diverse adult tissues. LY 411575 has been shown to promote goblet cell differentiation in mice intestine. Concurrently, LY411575 inhibits cell proliferation. LY411575 has also been observed to promote neural differentiation of mouse embryonic stem (ES) cells. [1][2]
