LY2228820 is aa novel and potent p38MAPK inhibitor (IC50 for p38αMAPK and p38βMAPK were 7 nM and 3 nM, respectively). LY2228820 enhances the toxicity of bortezomib against MM cells and modulates the cytokine network in BM microenvironment in vitro. Besides its effects modulating osteoclast formation in vitro., LY2228820 showed activity to inhibit in vivo osteoclastogenesis in a SCID-hu MM model. These results suggest that p38MAPK inhibition by LY2228820 represents a promising novel targeted therapy to improve the outcome of MM patients, by both reducing skeletal events and enhancing cytotoxicity of bortezomib.[1][2]
