MDM-2 is a key negative regulator of p53 that binds to and targets p53 for proteasomal degradation. Overexpression of MDM-2 in some tumors leads to inactivation of p53 thereby allowing tumor cells to escape p53-induced apotposis. p53 Activator III, RITA, also referred to as NSC 652287, binds to MDM2 (Kd = 1.5 nM), disrupting the MDM2-p53 complex and subsequently activating p53 and inducing apoptosis.
